A highly articulate, erudite and intelligent businessman and mastermind, "Red" Reddington, has allegedly been on the "10 Most Wanted List" of various U.S. law enforcement agencies for over 20 years. The legend is that Red is as elusive as he is clever, controlling a labyrinth of creative enterprises, coupled with uncanny ability to gather and finesse information at the drop of a hat. On the first day at FBI for a new female profiler fresh out of Quantico, Red offers to bandy wits with the FBI. Red promises to deliver various criminals and plots previously unknown to any branch of law enforcement... and all Red requests in return is to choose his muse.

Raymond "Red" Reddington, en av FBI's mest efterlysta brottslingar, kapitulerar på FBI:s högkvarter i Washington DC. Han hävdar att FBI och han har ett gemensamt intresse av att bli av med farliga brottslingar och terrorister, och att han har tillgång till ovärderlig information, men att han enbart kommer att tala med Elizabeth Keen,en nykläckt FBI-profilerare. Efter att FBI använt Reddingtons information för att hitta en terrorist, avslöjar Reddington att denna terrorist är bara den första av många brottslingar som han kommer att hjälpa dem att neutralisera. Under sin kriminella karriär har han gjort en lista över globala brottslingar som han tror är riktigt farliga för samhället, varav de flesta är okända för FBI.
